چکیده

LGK974 is a potent Wingless-related integration site inhibitor at Phase II of clinical trial. However, little known about its role and the underlying mechanisms in human hepatocellular carcinoma. 3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide colony formation assays were used to determine cytotoxicity on liver cancer cell lines. Flow cytometry assay was performed examine apoptosis induced by alone or combination with tumor necrosis factor- related apoptosis-inducing ligand. Western blot evaluate protein expression profiling caspases, poly adenosine diphosphate-ribose polymerase B-cell lymphoma 2 family, as well activation signaling pathways. At present study, we demonstrated that can suppress carcinoma proliferation inducing formation. In addition, effectively inhibited 2, while elevated homologous antagonist/killer, associated agonist death like 11 cells. More interestingly, it observed obviously enhances factor-related ligand apoptosis. Our data indicated not only site/beta catenin pathway, but also inactivated nuclear factor kappa light chain enhancer activated B cells pathway. We pathway using small interfering RNA against polypeptide gene B-cells alpha, which attenuated sensitization effects line, HepG2 Altogether, these presented this study illustrated potentiated lethality via suppressing

Monascin ameliorate inflammation in the lipopolysaccharide-induced BV-2 microglial cells via suppressing the NF-κB/p65 pathway

Objective(s): The pathophysiology of neurodegenerative diseases is complicated, in which inflammatory reactions play a vital role. Microglia cells activation, an essential process of neuroinflammation, can produce neurotoxic molecules and neurotrophic factors, which aggravate inflammation and neuronal injury. Monascin, a major component of red yeast rice, is an azaphil...

Nanocurcumin-Mediated Down-Regulation of Telomerase Via Stimulating TGFβ1 Signaling Pathway in Hepatocellular Carcinoma Cells

Background: Curcumin, extracted from turmeric, represents enormous potential to serve as an anticancer agent. Telomerase is viewed as a prominent molecular target of curcumin, and transforming growth factor-β1 (TGFβ1) has proven to be a major inhibitory signaling pathway for telomerase activity. In the current study, we aimed to explore suppressive effects of nanocurcumin on telomeras...

Angiopoietin-like protein 8 (betatrophin) may inhibit hepatocellular carcinoma through suppressing of the Wnt signaling pathway

Objective(s): Hepatocellular carcinoma (HCC) is one of the leading fatal neoplasms and the most common primary liver malignancy worldwide. Peptide hormone ANGPTL8 (betatrophin) may act as an important regulator in HCC development through the Wnt/β-catenin pathway. We aimed to evaluate the effects of recombinant ANGPTL8 on Wnt/β-catenin signaling in human liver carcinom...

ShDcR3 sensitizes TRAIL-resistant HCC cells by inducing caspase-dependent apoptosis while suppressing NF-κB dependent cFLIPL expression

Evidence has shown that most hepatocellular carcinoma (HCC) cells are resistant to tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL)-mediated apoptosis. However, the molecular mechanisms underlying TRAIL-mediated apoptosis resistance are not well understood. In this study, we reported that downregulation of Decoy receptor 3 (DcR3) expression by lentiviral vectors carrying sh...

AGK enhances angiogenesis and inhibits apoptosis via activation of the NF-κB signaling pathway in hepatocellular carcinoma

High levels of angiogenesis and resistance to apoptosis are major clinical features of hepatocellular carcinoma (HCC), a lethal disease with a high incidence worldwide. However, the precise mechanisms underlying these malignant properties remain unclear. Here, we demonstrated that acylglycerol kinase (AGK) is markedly overexpressed in HCC cell lines and clinical tissues. Immunohistochemical ana...
